Bacteriostatic Water for Peptide Reconstitution: What It Is and Why It Matters

Every lyophilized peptide must be reconstituted before research use, and the choice of diluent directly affects compound stability and usable life. The research standard is bacteriostatic water — and the reasons are practical, not arbitrary.
What Bacteriostatic Water Actually Is
Bacteriostatic water is sterile water containing 0.9% benzyl alcohol. The benzyl alcohol acts as a bacteriostatic agent — it inhibits the growth of bacteria that could otherwise contaminate a vial across multiple access events. This is the critical distinction from plain sterile water:
- Sterile water: sterile at the moment of first use, but offers no ongoing protection. Suitable only for single-use applications.
- Bacteriostatic water: suppresses microbial growth for the life of the vial — typically up to 28 days after first puncture — supporting the multi-access pattern typical of research protocols.
Why This Determines Peptide Usable Life
A reconstituted peptide vial is accessed repeatedly over days or weeks. Each access is a potential contamination event. Reconstituting with bacteriostatic water is what makes the standard 28-day refrigerated stability window achievable; with plain sterile water, contamination risk accumulates with every access regardless of refrigeration.
Proper Technique
- Swab both vial stoppers with alcohol and allow to dry
- Draw the desired volume of bacteriostatic water
- Inject slowly down the inner vial wall — never directly onto the lyophilized cake
- Swirl gently to dissolve; never shake
- Refrigerate at 2-8°C and observe the compound-specific stability window
